Outline of Research at the Start

細胞内は、分子クラウディング状態とよばれる高分子が300-400 mg/mLという高濃度で存在する状態にあり、タンパク質間やタンパク質・DNA間での相互作用における熱力学的状態や反応速度は、希薄溶液であるin vitroにおける状態とは大きく異なることが知られている。本研究では、細胞が細胞核よりも狭い数μmの間隙を移動中、細胞核膜が破裂して細胞核内包物が細胞質に放出される現象を利用して、細胞核内と細胞質内の分子夾雑環境の差異を定量的に評価する方法・デバイスを構築する。

Outline of Annual Research Achievements

真核細胞の細胞核内は、ヘテロクロマチンのように酵素反応のために酵素が侵入するのを妨げられたり、光の進行をも阻害されたりするほど超高密度の領域が存在している。このような分子クラウディング環境の内でも密度の局在分布があることは周知の事実であったが、これを時空間的に定量的に扱い、生体分子相互作用解析結果などへ適用した例はない。これまでの「均一な」分子夾雑環境からさらに踏み込んで分子夾雑の生命化学を理解するためには、「不均一な」分子夾雑環境を定量的に計測する方法論が必要不可欠である。そこで本研究では、細胞が細胞核よりも狭い数μmの間隙を移動中、細胞核膜が破裂して細胞核内包物が細胞質に放出される現象を利用して、細胞核内と細胞質内の分子夾雑環境の差異を定量的に評価する方法・デバイスを構築することを目的とした。
本年度は、細胞が狭窄部分を通過する際に破裂した細胞核が自己修復される前に細胞を回収する方法を検討するとともに、細胞核内包物と細胞質内包物が混ざった状態の細胞質成分をナノワイヤなどの極微細構造で取り出す方法を検討し、細胞核内の情報ができるだけ反映された状態で酵素活性の測定系へと持っていける系を構築してきた。その結果、単一細胞・単一細胞核レベルで細胞を解析するためのデバイスフォーマットの作製に成功するとともに、狭窄部分での細胞通過時間が細胞核の状態に大きく依存すること、また、細胞破裂に伴って細胞質内包物の多くがデバイス表面に吸着されることを明かとした。これらの研究成果により、細胞核内と細胞質内の分子夾雑環境を理解するための基盤技術を構築できた。
