Outline of Research at the Start

医薬品は化学物質であり、低分子であれば簡単な化学構造式で表現することができる。しかし、我々は化学構造式を見ただけで如何なる作用を発揮する物質であるかを当てるほど十分な知識を持ってはいない。本研究では、申請者らが構築してきた薬理作用予測モデルとリアルワールドデータを結合させることで、あらゆる化合物のヒトでの薬効・副作用の予測を可能とする。

Outline of Annual Research Achievements

化学構造のみから薬理作用を予測するモデルを開発するため、グラフ畳込みネットワークによる特徴量抽出法を導入し、ChEMBLに含まれる薬理作用データの予測性能を基にネットワーク構造の最適化を行った。前年度に構築した127種類の受容体/酵素/チャネルへの親和性を予測するモデルを拡充することで、薬理学的標的542種に対する作用予測プログラムを構築した。これにより、ChEMBL実アッセイデータの網羅率は83%に達した。構築した予測モデルの予測精度は標的タンパク質ドメインによらず高く(テストデータMAE < 0.6)、標的数の拡大によっても予測精度が維持されていることが示唆される。また、そのin vitroでの検証を進め、統合失調症治療薬の作用点である5-HT2A受容体に対する新規リガンドおよび気管支拡張薬の作用点であるβ2アドレナリン受容体に対する新規リガンドを得た。これらの結果は、構築した予測モデルの高い妥当性を示唆するものと考える。
次に、ヒトにおける副作用/薬効を因果関係を考慮して予測するため、医薬品処方と副作用発現の順序関係が明らかなヒト副作用ビッグデータであるJADERを用いて、薬理作用予測値から副作用発症を予測するモデルを構築した。その結果、SMQでグルーピングした疾患群の74%についてROC-AUC0.7以上の良好な予測モデルを構築できた。さらに、Explainable AI技術の一つを用いて予測モデルの内容を調査したところ、既報から明らかな標的タンパク質のみならず新規な抗うつ薬の創薬標的となる複数の標的タンパク質が見出された(投稿準備中)。
