Outline of Research at the Start

本研究では、質量分析装置を用いてリボソーム上で合成されている新生ポリペプチド鎖を大規模にプロファイルするテクノロジー(プロテオームリボソームプロファイリング法)を創出することを目的とする。さらに本法を用いて、従来の総タンパク質を対象としたプロテオーム解析では捉えることが困難であった摂動によって誘導される分オーダーの翻訳応答をモニターし、翻訳レベルでの迅速なタンパク質合成制御機構を明らかにする。

Outline of Annual Research Achievements

本研究では、質量分析装置を用いてリボソーム上で合成されている新生ポリペプチド鎖を大規模にプロファイルするテクノロジー(Uchiyama et al., bioRxiv 2021, pSNAP法: Puromycin- and SILAC-labeling based Nascent Polypeptidome Profilng)と(Uchiyama et al., J. Biochem. 2020, OPP-SILAC法)を開発した。さらに本法を用いて、従来の総タンパク質を対象としたプロテオーム解析では捉えることが困難であった摂動によって誘導される分オーダーの翻訳応答をモニターし、翻訳レベルでの迅速なタンパク質合成制御機構を明らかにした。具体的には、LPSで刺激したマクロファージ細胞の翻訳応答やマウス初代神経細胞の分化過程での翻訳応答を明らかにした。本法は従来よりも安価、簡便(化学標識やリボソームの精製を必要としない)、新生タンパク質と非特異タンパク質を区別することが可能といった特長を有する。今後は、開発した手法を用いてin vivoやex vivoでの翻訳応答を捉えることが可能な技術に拡張していく予定である。
