Outline of Research at the Start

植物幹細胞は多能性の維持において独自のゲノム制御システムを有している。植物幹細胞はDNA損傷に対して高感受性を示すが、細胞死に至るまでのシグナル応答は多くが未解明である。これまでの研究から、植物幹細胞が DNA 損傷を受けた場合、オーキシン発現量の低下に伴うクロマチン弛緩が生じることが明らかになりつつある。本研究では、動物細胞を植物研究のモデル生物として用い、幹細胞における DNA損傷発生、修復経路、そして細胞死までの分子機構を解明することで、動植物幹細胞ゲノム維持機構の共有性と独自性の統合的理解に迫る。

Outline of Annual Research Achievements

植物幹細胞は多能性の維持において独自のゲノム制御システムを有している。植物幹細はDNA損傷に対して高感受性を示すが、細胞死に至るまでのシグナル応答は多くが未解明である。これまでの研究から、植物幹細胞がDNA損傷を受けた場合、オーキシン発現量の低下に伴うクロマチン弛緩が生じることが明らかになりつつある。本研究では、動物細胞を植物研究のモデル生物として用い、幹細胞におけるDNA損傷発生、修復経路、そして細胞死までの分子機構を解明することで、動植物幹細胞ゲノム維持機構の共有性と独自性の統合的理解に迫る。本年度は、細胞の運命を左右する重篤なDNA損傷の一つであるDNA二本鎖切断(DSB)後の損傷応答に着目し、DSB誘導後のクロマチン構造変化とDSBマーカーの局在性を、超高解像度蛍光顕微鏡3D-SIMを用いて解析した。また、DSB発生時の核内クロマチン弛緩状態をモニターするため、部位特異的DSB誘導系であるAsiSIアッセイを構築し、ATAC-seqを行った。これらの結果から、DSB部位の近傍のみにおいてクロマチンが弛緩し、遠位では複数のヌクレオソームが構築するナノドメインの配置変化が起こることを見出した。さらに、DSB後のクロマチン構造変換に影響を及ぼす分子機構を解明すべく、DNA損傷後の遺伝子発現をRNA-seqによるトランスクリプトーム解析を行った結果、分化した細胞ではCAF-1,HP-1,KAP-1の顕著な低下が認められたことから、遺伝子発現レベルでのクロマチン構造制御が行われている可能性が示唆された。
