Outline of Research at the Start

人工骨髄の構築は申請者の最終的な研究目的のゴールであり、基礎研究や臨床研究の面において重要である。その理由として、人工骨髄システムを用いることで造血幹細胞から全ての血液細胞を分化誘導が可能であり、造血幹細胞を維持できる予想から新たな造血幹細胞バンクの基盤を創造することができる。また骨髄には間葉系幹細胞や他に様々な未熟な未分化細胞が存在することから移植細胞のソースとして応用可能であると考えられる。さらには人工骨髄をアクリルケースにいれた蟻の巣のような観察システムを構築することで正常細胞や白血病細胞の動態を明確にする。

Outline of Annual Research Achievements

重篤な血液疾患を根治するために造血幹細胞移植が行われているが,ドナー不足の問題は深刻である.この問題を解決する為に,造血幹細胞を生体外で増幅させる技術の開発が重要な課題となっている.造血幹細胞を生体外で増幅させるには,1)造血幹細胞の未分化状態を維持しうる骨髄全体および骨髄微小環境の構造,2)髄液の構成成分(栄養成分など),の詳細な理解が必須である.令和3年度は現在までに骨髄微小環境の構造やそれを構築している細胞,さらには髄液中の成分を明らかにした.さらに詳細な骨髄の構造,構成成分に関する情報を取得し,この情報を基に,マイクロ・ナノ工学を駆使することで造血幹細胞が増殖可能な骨髄微小環境を生体外で構築することを目指した.生体外骨髄微小環境の構築により,ドナーにたよらない造血幹細胞提供システムの開発や骨髄微小環境ごと患者の体内に移植するという新しい骨髄移植法の確立するとともに、造血幹細胞移植後の骨髄再構築への細胞動態を時系列的に解析した。
