Outline of Research at the Start

造血幹細胞が造血系を維持する機構を解明することは、他の臓器・細胞の恒常性の維持機構の解明にも応用でき、造血系の恒常性の破綻に起因する疾患の研究において基礎的な知見を与える。「細胞ダイバース」前期では、新しい造血幹細胞分化経路を含んだ分化モデルを定式化し、造血幹細胞移植の実験データから各種のパラメータの推定するアプローチを開発した。しかし、国内外を見渡しても、老化（エイジング）により、造血幹細胞の能力がどのように変化し、その結果として血液細胞動態がどの程度変容あるいは破綻するのかは、いまだ明らかにされていない。「細胞ダイバース」後期では、この老化に取り組む。

Outline of Annual Research Achievements

移植実験で得られたキメラ率のデータのうち、移植した１細胞がlong-term HSC（LT-HSC）およびintermediate-term HSC（IT-HSC）と分類されたものを用い、非線形混合効果モデルを用いて開発した数理モデルのパラメータ推定を行った。推定した若齢および加齢マウスの血液細胞系譜を特徴づけるパラメータから、若齢マウスでは骨髄球のみを産生するMyRPs（myeloid repopulating progenitors）からの赤血球および血小板への産生の寄与率が、HSCから骨髄球バイパスを経由しない場合より大きいことがわかった（図）。また、加齢による影響として、HSCの自己複製能が半減し、各細胞系譜への分化能が亢進していることも示された。なお、推定されたパラメータを用いて計算することで、加齢によって骨髄球の産生自体が増加し、加えて、骨髄球バイパスを経由する産生の割合が増加していることが示された。この結果は、多くの研究で示唆されている、加齢によって造血が骨髄球に偏る（myeloid shift）ことを、HSCの能力の変化のレベルと細胞分家系譜全体としての造血システムのレベルで説明できるのではないかと考えられる。さらに、移植時に初期の造血を補助するために１細胞と同時に移植される骨髄細胞（競合細胞）の中に含まれるHSCとMyRPの数も同時に推定することで、不明であった競合細胞の組成を明らかにするとともに、キメラ率への競合細胞の影響についても定量化することができた。将来的には、老化により、造血幹細胞の能力がどのように変化し、その結果として血液細胞動態がどの程度変容あるいは破綻するかが定量的に解明されると期待できる。
