Outline of Research at the Start

本研究では、独自に開発した膜タンパク質の分布を高分解能・高感度に可視化できるSDS凍結割断レプリカ標識法(SDS-FRL)を用いて、海馬シナプスにおけるAMPA型グルタミン酸受容体(AMPAR)のシナプス内発現分布とその神経活動に応じた変化を、光遺伝学的操作や種々の薬理学的処置を駆使して解析し、「AMPARのシナプス内発現密度を制御している機構がシナプスのメタ可塑性(可塑性発現能)の構造基盤であること」を単一シナプスレベルで証明すると共に、その分子機構や関連する神経修飾系を明らかにして、「脳の記憶機構の本質」に迫る。

Outline of Annual Research Achievements

神経活動依存的なシナプスの伝達効率の変化(シナプス可塑性)は学習・記憶の基盤となる細胞レベルの現象として、その分子メカニズムが活発に研究され、中枢の興奮性シナプス伝達の効率がシナプス後膜上に発現するAMPA型グルタミン酸受容体(AMPAR)の数に依存することが示されている。しかし、シナプス可塑性の起こりやすさ(メタ可塑性)のメカニズムについては、殆ど明らかにされていない。そこで本研究ではシナプス内AMPAR密度がメタ可塑性の構造実体であることを証明することを目的とし、①受容体局在解析法の高感度高分解能化、②-1光遺伝学的手法を用いた神経活動とシナプス内受容体発現様式の関連性の解析、②-2 遺伝子欠損マウスの受容体分布の解明、②-3 行動薬理学的条件下でのシナプス内分布変化の解明の4つの実験を計画した。
このうち、令和3年度は②-2~3を実施した。②-2では、N-cadherin及びNeuroligin-1-P89Lマウスのシナプス内AMPAR発現密度と分布が野生型マウスと有意な差を示さず、これら分子がシナプス内AMPAR発現に直接関与していないことを示唆する結果を得た。②-3では令和4年5月時点で、標識レプリカの作製まで完了し、今後も引き続き解析を進め論文化する予定である。
また、本年度は領域内の共同研究にもこれまで以上に時間を割き、名古屋大学山中研究室で解析中のMCHニューロンのシナプス構造について免疫電子顕微鏡を用いた解析を行った。その結果、MCHニューロンの海馬内錐体細胞に対するシナプス結合は、I型とII型の両方の構造様式を示すことを明らかにした。
