Outline of Research at the Start

幹細胞は、発生初期には自己複製によりその数を増やし、あるタイミングになると分化細胞を産生する。すなわち、複製・維持されている非ゲノム情報を、あるタイミングで変換することで、適切な組織を構築できる。本研究では、神経幹細胞が自己複製を繰り返す増殖期からニューロン分化を開始する時期にかけて非ゲノム情報であるオープンクロマチン領域をどのように複製・維持あるいは変換することでその運命を制御しているのかを明らかにする。

Outline of Annual Research Achievements

本年度は、神経幹細胞の時期依存的な変化を担う非ゲノム情報の探索を実施した。
増殖期、ニューロン分化期、そしてグリア分化期の神経幹細胞を採取し、様々なヒストン修飾に対するCUT&Tagを実施した。そして、その非ゲノム情報を用いて神経幹細胞の状態変化を調べたところ、不活性ヒストン修飾の情報を用いることで、1. 増殖期からグリア分化期まで時期依存的に徐々に変化していく非ゲノム情報と、2. ニューロン分化期におけるニューロン分化能と相関する非ゲノム情報に分離できることがわかった。
本研究は、非ゲノム情報を用いて神経幹細胞の状態を予測できることを示している。
