Research Abstract

本研究課題では,ボトムアップおよびトップダウン方式により作成した金ナノ構造体をベースとした光-分子強結合反応場を用いて,近赤外蛍光の強度に及ぼす影響について検討し,蛍光量子収率の低い色素分子の蛍光増強手段としての可能性、具体化、定量化の検討を目的とした.光-分子強結合場で蛍光放射速度定数を制御できるかについての検討も試みた.今年度は金属ナノ構造体を利用した近赤外励起蛍光の検討および励起波長および偏光依存性を中心に検討を行った.
電子ビームリソグラフィーにより作成された金ナノアレイを用いて近赤外光励起による蛍光の検討を行った.疎水化DNAを媒体として,近赤外蛍光色素IR780の蛍光増強に関する検討を行った.金ナノアレイの存在によって,14倍程度の蛍光増強(金属のある領域の面積で規格化)を達成した.次に,蛍光増強度の励起波長依存性を検討したところ,金ナノアレイのプラズモン吸収(消衰)スペクトルに対応した蛍光増強を示した.一方,IR780の蛍光スペクトルとプラズモン吸収とが重なる場合,ほとんど増強を示さなかった.これは,色素の励起状態から金ナノアレイへの励起エネルギー移動効率の増大によって蛍光消光が生じたためと考えられる.また,s偏光励起に比べ,p偏光励起の方が大きな増強効果が得られた,この事実は,蛍光増強が金ナノアレイの光学特性に依存した現象であることを示している.
局所電場増強がさらに大きいと期待される金ナノブロックのダイマー構造における増強効果の検証についても予備的な知見を得ている。このように本成果は、規則構造を有する金属ナノ構造体を利用した近赤外蛍光増強可能な光-分子強結合反応場の創成に重要な寄与が期待できる.
