Research Abstract

H21年度は以下の2点についてそれぞれ成果を得た。以下に列記する。
1.ウイルス感染モデルを用いたRNA応答の解析:当研究室にマウスモデルが存在する麻疹、ポリオ、VSVを用いて宿主のウイルスRNA応答を解析した。麻疹の野外株はIFNを誘導しないが、馴化株はRIG-I/MDA5経路でtype I IFNを誘導する。この誘導機構はDI RNAによるもの(Shingai et al., J Immunol 2007)の他、V蛋白の変異(MDA5を抑制できない)もあることが判明した(Takaki et al.,未発表)。ポリオウイルスはPVR+マウスに感染し、IFN誘導を誘起する。この原因はマクロファージが感染細胞を貪食してTLR3/TICAM-1経路を活性化することが主因と判明した(Oshiumi et al.,未発表)。IPS-1 KOマウスでもHCVの場合と同様、IFN誘導は十分に起きた。これらの結果はIPS-1経路のみがウイルス感染の防御機構を担うのではなく、感染形態に合わせて複数の抗ウイルスIFN誘導経路が活性化することを証明する。樹状細胞におけるTICAM-1経路はIFN以外にNKも活性化するので、感染免疫への関与を検討中である。
2.合成RNA誘導体による樹状細胞を介した免疫活性化:PolyI:Cはcytokine stormなど有害事象を誘起するが、これはIPS-1経路(RIG-I/MDA5)とTICAM-1経路を両方活性化するためである。後者は樹状細胞など限られた発現を示し、免疫系を活性化するのでTICAM-1指向性のRNA誘導体を作製した(特許申請中)。エンドソーム標的型RNA誘導体がこの目的に合致し、mDCのThR3を選択活性化出来る事が判明した。毒性が低くmDCを成熟化させるRNA誘導体をこの系で抽出する事を試みている。
