Research Abstract

先に、C型レクチンSIGNR1が病原性C.albicans表面のα-mannanを介してこれを認識し、細胞のTNF-〈産生およびoxidative burstを昂進すること等を細胞株にて示した。今年度は、生体におけるSIGNR1の働きを検討した。
始めに、マウスに抗SIGNR1抗体を投与すると腹腔内マクロファージ(rpMφ)上のSIGNR1が消失することを確認した。次に当該マウスの腹腔内にzymosanを投与し一定時間後に腹腔内細胞を取り出し解析したところ、抗体処理によってrpMφのzymosan捕捉と続くTNF-α産生が有意に低下した。TLR2KOマウスでは、zymosan投与によるTNF-α産生が大きく低下したことから、先のin vitroの結果と同じく、生体においてもSINGR1はTLR2を介してrpMφのTNF-α産生を昂進することが示された。一方で、熱処理C.albicansのrpMφによるoxidative burst誘導における抗SIGNR1抗体処理の影響を同様に検討したところ、抗体処理によりrpMφのoxidative burstが有意に低下した。先に、細胞株で見られたSIGNR1とDectin-1の結合をrpMφにおいても検討したが確認されなかった。しかしながら、熱処理C.albieans刺激の後に両者の共局在および結合が確認され、更に、Dectin-1が認識しにくい生菌C.albieansをrpMφに感染させたところ、菌体周囲にSIGNR1に加えてDectin-1の局在が見られた。先に、SIGNR1はDectin-1/Sykを介してoxidative burstを惹起することを報告していることから、SIGNR1は生菌体の取り込みと共にDectin-1をその周囲に導き効果的なoxidative burst誘導を可能にしているものと思われる。
