Research Abstract

本研究は細胞内たんぱく質分解現象であるオートファジーが哺乳類の上皮系の細胞に侵入したサルモネラの増殖制御に関与する分子機構を明らかにする目的で行った。サルモネラは腸皮細胞など上皮系細胞へと侵入後、サルモネラ含有小胞と呼ばれる膜構造体で緩やかな増殖をするが、オートファジーの欠損したマウス胚性線維芽細胞ではそれに加えて細胞質中で爆発的な増殖が見られる。オートファジー関連タンパク質LC3は侵入したサルモネラの一部の周囲にリクルートされてくるが、それはLC3のユビキチン脂質化反応に依存していた。ところが興味深いことに、やはりオートファジーに関わるAtg9L1の欠損したマウス胚性線維芽細胞では、サルモネラの増殖抑制が見られないにもかかわらずLC3がサルモネラの周囲に効率よくリクルートされてきた。この細胞ではオートファジーによる2重のも開く構造が見られなかく、サルモネラを囲む一重の膜構造にLC3がリクルートされることがわかった。このことよりサルモネラオートファジーにおいてLC3はオートファゴソーム様の膜構造ができなくても、サルモネラの周囲に特異的にリクルートされてくる機構が存在することが明らかになった。
