原虫感染時のサイトカインによる炎症誘導機構の解析とその制御に関する研究

Research Abstract

本研究は、原虫感染に伴う免疫・炎症病変の誘導機構、およびその生理的・病理的意義を明らかにすることを目的とした。本研究では、1)自然免疫細胞における、CBM複合体を介した新しい炎症性サイトカイン産生機構の解析とその意義の解明、2)自然免疫細胞が産生するサイトカインによる炎症誘導が獲得免疫の成立に与える影響、3)獲得免疫において産生される、IL-17などの炎症誘導性サイトカインやIL-27などの免疫抑制性サイトカインが、自然免疫・獲得免疫両者に与える影響、を検討する。1)2)に関して、原虫Leishmania majorがCBM複合体を介して自然免疫と引き続く獲得免疫の誘導を行うことを明らかにした。現在、認識する受容体やそのリガンドの同定を試みている。また、Trypanosoma cruz感染においても本経路の重要性を明らかにしつつある。3)に関しては、既にIL-27による角の炎症抑制作用やIL-17による炎症誘導作用を明らかにしてきた。本研究を通じて、原虫感染防御免疫における、Toll様受容体を介さない、新規の経路による自然免疫活性化機構が明らかにされつつあり、さらに、この経路が引き続く獲得免疫の誘導に及ぼす効果も明らかにされてきた。これらは、ウイルスや細菌とは異なる、真核病原体である原虫や真菌に対する免疫の成立経路を明らかにする上で重要な知見であり、既知の自然免疫経路、および既知のTh1細胞によるものとは異なる経路による原虫(および真菌などの真核病原体)に対する感染防御機構の解明につながる新しい研究テーマとなると期待される。
