細菌感染におけるM細胞の生理機能と生物学的重要性の解析

Research Abstract

我々はこれまで、FAEのトランスクリプトーム解析データを手がかりに、M細胞に発現する病原体取り込み受容体の探索を行い、2種のGPIアンカー型蛋白質Glycoprotein 2(GP2)およびCellular prion protein(PrP^C)を同定した。昨年度までに、GP2は、Escherichia coliやSalmonella TyphimuriumなどのI型線毛と選択的に結合し、これらの細菌のM細胞内への取り込みを促進することを報告した。さらに経口免疫実験の結果から、GP2依存的な細菌の取り込みは、抗原特異的な粘膜免疫応答の発動においても重要であることを明らかにしてきた。
今年度はPrP^Cの分子機能について解析を行った。PrP^Cは生体内に広く発現しているものの、その発現量は組織、細胞によって大きく異なっている。マウスの腸管上皮層ではM細胞に特に強い発現が観察された。共焦点顕微鏡観察の結果から、PrP^CはM細胞の管腔側細胞膜に局在しており、抗原取り込み受容体として機能することが示唆された。PrP^Cリコンビナントタンパク質を用いたin vitro結合試験の結果から、PrP^Cは人獣共通感染菌の一つであるBrucella abortusと結合した。腸管ループ内にブルセラ菌を注入したところ、ブルセラ菌はM細胞に選択的に取り込まれ、その際PrP^Cとの共局在を示した。さらにPrP^C欠損マウスでは、ブルセラ菌のパイエル板や腸間膜リンパ節への取り込みは大幅に減少した。以上の結果から、M細胞はブルセラ菌経口感染時の経路の一つであり、その際にはPrP^Cが侵入受容体として機能することが示唆される。
