生体モーター運動活性の局所的熱変調

Research Abstract

レーザー光による光刺激を用いてべん毛モーターの回転速度を可逆的にかつ局所的に操作することで、生体内分子機能をリアルタイムで制御する新しいツールを開発することを目指し、平成22年度は以下を行った。
1)光刺激用レーザーを導入した計測系の評価:昨年構築した光刺激用レーザーを組み入れた計測系について、温度変化と細胞へのダメージを検討した。まず、プローブを用いないで細胞そのものへYAGレーザーを集光して回転運動を観察すると、YAGレーザーの最大強度2Wでも顕著な変化が見られなかったのに対して、ポリスチレンビーズ(直径1um)をべん毛に結合させて、YAGレーザーを与えると、照射時に回転速度の変化や回転の停止が観察された。このようなレーザー照射の影響は、可逆的に引き起こされ、ビーズの直径を半減させるとほとんど見られなくなり、プローブの大きさや熱的特性に依存することが示唆された。
2)局所熱刺激によるべん毛モーターの運動応答の計測:構築した計測系を用いて、レーザーによる局所的光刺激下でのべん毛モーターの運動性を計測した。ポリスチレンビーズより、光-熱変換効率の高いプローブとして、金コロイド粒子(直径250nm)での計測を行った。ポリスチレンビーズでの速度変化4%に対して、金コロイド粒子を用いた場合、速度は可逆的に50%以上高速化することに成功した。この時の温度変化は、金粒コロイド粒子表面で200℃程度に対して、モーター自身の温度上昇は2℃程度と考えられる。本研究の結果は、生体分子の温度を、プローブの直接的結合によって100℃以上の広温度域で制御できる可能性と、プローブの間接的導入によって、温度変化をごく局所領域にのみ制御できる可能性を示した。
