Research Abstract

全くユニークなメカニズムである,マイコプラズマの滑走運動における,"あし"の動きを明らかにするために,以下の研究を行った.(1)活性のある滑走装置を単離し,ATP加水分解サイクルーガラス結合一動き,の関係を明らかにした,(2)低濃度の界面活性剤で処理することで細長く伸びた細胞の滑走運動を解析することにより,力発生の最小単位を見積もった.(3)シアル酸オリゴ糖や,ライブラリー抗体により細胞が固形物表面から解離する直前の動きを解析することで,"あし"が協同的に動いていることを示唆した.(4)滑走装置のタンパク質に結合して,滑走を阻害するライブラリー抗体を40個取得し,滑走時における力の伝達を明らかにした.また,これらの抗体を用いることで,滑走タンパク質各部位を蛍光ラベルすることに成功した.(5)滑走装置を構成する3つのタンパク質,Gli123, Gli349, Gli521のネガティブ染色電子顕微鏡法による構造解析を進めた.また,これらのタンパク質の液中における挙動を高速AFMによって明らかにした.
