Research Abstract

26Sプロテアソームは,高等植物が持つ優れた環境適応能力を説明するための分子機構候補として注目を浴びている。申請者らは,モデル植物シロイヌナズナを用いて,19Sプロテアソームを構成するサブユニットRPT2aの機能欠損変異体(rpt2a)において,葉器官等が巨大化する現象を発見した。その後の研究により,この原因がエンドリデュプリケーションという細胞周期異常に由来する細胞サイズ増大に起因することを明らかにした。また逆遺伝学的解析から,rpt2a変異体において過剰な遺伝子サイレンシングが生じることを発見した。
今年度は、RPT2aを介した遺伝子サイレンシング機構をより詳細に解析した。RPT2a遺伝子欠損によるサイレンシングは,内生だけでなく外生遺伝子にも影響することが明らかとなった。また,bisulfite法を用いた解析より,このサイレンシングにDNAのメチル化が関与することが示された。次に,核プロテオミクスによる,RPT2a新規相互作用因子の単離を試みた。その結果,遺伝子サイレンシングに関与する可能性のあるタンパク質Xを同定することに成功した。新規相互作用因子Xの詳細な解析によって,RPT2aの作用機序の解明が期待される。
また,RPT2aはエンドリデュプリケーションならびに遺伝子サイレンシングを負に制御することから,これら両現象の関連性について,遺伝学的解析を行った。その結果,エンドリデュプリケーションが遺伝子サイレンシングを促進させる可能性を明らかとした。
