Research Abstract

本研究ではI-CLiPの中でも特にゴルジ体に局在することが明らかになっているS2PとRhomboidファミリーのRHBDL1とRHBDL2、またS2Pと協調して機能しているS1Pに注目して、これらの解析を行った。昨年度に引き続いてHeLa細胞cDNAライブラリ、ヒト神経芽腫細胞株であるSH-SY5Y cDNAライブラリなどを用いてRHBDL1のクローニングの試みを続けたが、相当のクローンを得ることができなかった。従って、RHBDL1のクローニングには、他の適当なライブラリを探索する必要がある事が明らかとなった。昨年度、cDNAを取得できたS1P,S2P,RHBDL2について、酵母ツーハイブリッドシステムを用いたスクリーニングを行ったが、バックグラウンドが高く、擬陽性の候補が多数得られたため、残念ながら特異的なシグナルを得る候補遺伝子の特定に至ることはできなかった。ゴルジ体と小胞体に局在する機能未知の5回膜貫通タンパク質であり、I-CLipと特異的相互作用が期待されるYIPFファミリーとの複合体形成の解析を行った。S1P,S2P発現細胞の細胞溶解液をネイティブPAGE法、および免疫プルダウン法で解析したところ、YIPFとS1P,S2Pは、ネイティブPAGE法で別の分子量の複合体として検出され、また、プルダウン法で両者の共分離を検出することはできず、残念ながら有意な相互作用を示唆する実験結果を得る事ができなかった。
