精子走化性における誘引物質のセンサー機構と鞭毛運動制御のメカニズム

Research Abstract

多くの生物の受精時において、精子は卵へ走化性運動を示す。この現象は、卵を中心とした濃度勾配を形成している誘引物質を関知する精子上のセンサー分子が運動を制御していると考えられるが、分子機構に関してはまだごく一部しか解っていない。研究代表者の吉田らは、原索動物カタユウレイボヤの精子活性化・誘引物質が新奇の硫酸化ステロイドであることを明らかにし、SAAFと命名した。しかし我々の研究も含め、精子が誘引物質を認識するセンサーメカニズムについてはまだほとんど何も解っていない。そこで、これまでの知見及び構築した実験手法を用いて、受精時に見られる精子走化性現象において、精子がどのように空間勾配を形成する誘引物質を感知し、運動機能を変化させているかを解明することを目的とする。実験材料としては、既に誘引物質が我々の手で同定され、さらにゲノム解読も既に終了している原索動物カタユウレイボヤを用いる。今年度は特に精子誘引物質SAAFをセンサーする分子である精子上のSAAF受容体の同定に取り組んだ。まず候補分子として見つかっている370kDaタンパク質(P370)の精製法の改善行い、多量に精製することに成功した。そして、p370の同定をMALDI TOF-MSによって同定を行ったところ、細胞膜型カルシウムポンプであるPMCAであることを明らかにし、カタユウレイボヤPMCA(CiPMCA)を同定した。また、CiPMCA遺伝子はゲノム上1つで哺乳類と相同性が高く、少なくとも2つのsplicing variantがあった。このうち精子で高発現しているvariantを同定した。
