Research Abstract

Anabaena Sensory Rhodopsin(ASR)は、光反応過程、光誘起構造変化、情報伝達タンパク質(ASRT)との相互作用が、これまで研究されてきた古細菌型ロドプシンとまったく異なる新奇な光センサー蛋白質である。本研究では、研究代表者が最適化してきた液中で計測可能な全反射赤外分光計測を駆使することにより、生理的条件に近い温度、pH、塩濃度環境下での計測を実現し、ASRと情報伝達タンパク質(ASRT)の光情報伝達過程に伴うタンパク質の構造変化や水素結合変化を明らかにする。さらに、内向きプロトンポンプ活性を示すD217E変異体の特性を生かし、そのC末端にK^+チャネルであるKcsAを遺伝子工学的手法により連結させ、光応答性K^+チャネルを創成する。
平成22年度は以下のような成果を得た。1)ASRとKcsAとを結合したコンストラクトを作製し、大腸菌で発現させ、界面活性剤(DDM)で可溶化し、Ni-NTAカラムで精製を行った。各ドメインが適切にフォールディングされ活性を保持していることを、ASRドメインについては可視吸収分光法、KcsAドメインについては全反射赤外分光計測によって確認し、光応答性K^+チャネルの端緒となる結果を得た。2)セルセンサーの代表例である視覚のロドプシンの熱雑音の分子機構について、Thr118のH/D交換反応を指標にした解析を行い、11-cisレチナールの熱異性化が蛋白質の構造揺らぎによって許容されることを明らかにした(Lorentz-Fonfria, et al.J.Am.Chem.Soc.)。3)AO3班の七田芳則教授との共同研究として、G蛋白質を活性化するキメラ蛋白質の作成に成功した(Nakatsuma, et al.Biophys.J.in press)
