Research Abstract

我々はこれまで、可視化細胞と画像解析により、細胞分裂、分化形態形成について解析を行ってきた。具体的には、細胞内構造を複数の蛍光タンパク質や生体染色試薬により多重可視化したタバコBY-2細胞およびシロイヌナズナ植物体を用い、メリステムやモデル細胞の細胞分裂・分化における各細胞内構造の動態を生細胞で追跡してきた。本年度は、1)GFP-ABD2によりアクチン繊維を可視化したタバコBY-2の形質転換細胞にアクチン束化誘導剤であるTIBAおよびJasplakinolide(Jasp)を処理すると、表層および紡錘体周囲のアクチン繊維パターンが変化し、紡錘体および細胞板が著しく傾斜することなどから、細胞分裂期のアクチン繊維パターンが紡錘体の向きを制御する機構について解析した。2)多くの双子葉植物の葉表皮細胞はジグゾーパズルのように複雑な形状をとるが、ゴルジ体トランス層膜に局在するSialyl Transferase(ST)と蛍光タンパク質mRFPの融合タンパク質ST-mRFPを発現するシロイヌナズナの葉表皮組織において、ST-mRFPがアポプラストに局在し、細胞の成熟に伴って、表皮細胞では湾曲部に、孔辺細胞では細胞端部において顕著に見られたことから、多胞体などの細胞内構造を介して細胞質成分をアポプラストへ輸送する経路が、葉の表皮細胞の形態形成に関与する可能性を示した。3)ホンモンジゴケの銅耐性および応答性について検討し、ホンモンジゴケ原糸体における無性芽形成への銅添加の影響を調べた結果、添加した銅濃度依存的に無性芽形成が抑制されることから、銅によるホンモンジゴケの細胞分化制御機構の存在を示した。
