Research Abstract

植物細胞内に多数存在するオルガネラは,動物とは異なる多様でユニークな機能を持つことが明らかとなりつつある.これら植物特有の多様なオルガネラ機能は,そこに局在するタンパク質の組成や分子間相互作用の多様性を反映したものであると考えられるが,その発現機構は依然未解明の部分が多く残されている.本研究においては,バイオイメージングを主要な手段として,植物のオルガネラ機能のさらなる解明と,その多様性獲得機構を明らかにすることを目指している.平成22年度においては,植物固有のRAB5であるARA6のエフェクター候補分子,PUF2について,詳細な機能解析を行うとともに,その細胞内局在を様々なオルガネラマーカーを用いて観察した.その結果,PUF2がRAB5 GEFであるVPS9aのエンドソームへの局在化を促進すること,ARA6よりもむしろ,保存型RAB5であるARA7とより高い共局在性を示すことなどが明らかとなった.また,変異体を用いた遺伝学的解析においても,PUF2が保存型RAB5と強い遺伝学的相互作用を示すことが明らかとなった.これらの結果から,PUF2はVPS9aを介し,植物特異的なRAB5と保存型RAB5の機能を統御する因子であることが明らかとなった.ARA6は,RAB5に最も高い類似性を示しながら,通常のRAB GTPaseとは異なる構造を持つ陸上植物に特異的なRAB GTPaseである.このことから,植物が,進化の過程において,独自のRAB5分子を獲得し,それに伴い新たな膜交通経路を開拓してきたことが示唆される.
