乾燥ストレスからの回復過程におけるヒストン修飾を介した転写制御機構の解析

Research Abstract

シロイヌナズナを用いて、乾燥ストレス処理とその回復過程(再吸水処理)での遺伝子活性化から抑制までの過程におけるクロマチン動態の経時的変化について、クロマチン免疫沈降-定量PCR法(ChIP-qPCR法)を用いた解析を行っている。いくつかの乾燥ストレス誘導性遺伝子領域(RD20、RD29A、COR15A)において、RNA pol IIの結合は、再吸水処理により速やかに発現遺伝子領域から脱落することが明らかとなった。また同様に、転写活性化状態のマーカーであるヒストンH3Lys9のアセチル化はRD20およびRD29A領域において、再吸水処理により速やかに減少することがわかった。これに対して、COR15A領域におけるH3Lys9アセチル化は、緩やかに減少することがわかった。このことから、遺伝子発現抑制に伴うH3Lys9アセチル化変動(減少)はそれぞれの遺伝子領域に依存して異なることが示唆された。また興味深いことに、これら全ての遺伝子領域において、もう一つの転写活性化マーカーであるヒストンH3Lys4部位のトリメチル化は、転写抑制後にもある程度維持されることがわかった。一方、転写抑制状態のマーカーであるH3Lys27のトリメチル化量は、これら全ての遺伝子領域において、遺伝子発現応答の変動(抑制状態)とよく相関していることがわかった。
また、遺伝子変異株を用いた発現解析の結果から、いくつかの乾燥ストレス誘導性遺伝子の活性化に関わるヒストンアセチル化酵素の候補を同定した。
