精原細胞の減数分裂と精子形成パキテン期通過に必須の核内因子Dmrt7の機能解明

Research Abstract

Dmrt7タンパク質は生後12日から精巣内に発現し始め、精母細胞の核内に局在することを確認した。核内では、染色体様の構造と、XY bodyの両方に存在していることを、Scp3抗体とDmrt7抗体、γH2AX抗体とDmrt7抗体を用いた二重染色により明らかにした。
Dmrt7欠損マウスの解析では、HE染色により、精母細胞の脱落、多核巨細胞の出現を確認した。PAS染色からは、Dmrt7欠損マウスの精子形成異常が形態的に観察され始めるのは生後14日目からであることが明らかとなった。マーカー解析の結果、Dmrt7が欠損した精巣では、パキテン期中期のマーカー(Hoxa4)が若干発現低下しており、パキテン期後期のマーカー(Cdc25c)の発現は完全に低下していた。以上の結果より、Dmrt7はパキテシ期の通過に必須であり、減数分裂特異的なイベントに関与していると考えられる。
第一減数分裂前期には、減数分裂を特徴付ける様々なイベントが観察される。それらのイベントは減数分裂に必須であり、正常に機能しない場合は精子形成停止の主な原因となりうるp免疫染色と遺伝子発現解析の結果、Dmrt7欠損マウスの精巣では、相同染色体の対合、XYbody形成、MSCIは何れも正常である可能性が高いことが示唆された。しかし、MSCIのバックアップとして発現する遺伝子の一つであるPgk2の発現は野生型マウスと比較して約18%に低下していた。また、非LTR型のレトロトランスポゾンであるLINE1と、LTR型のLAPの発現が約2倍に上昇していることがqRT-PCRの結果明らかとなった。バックアップ遺伝子の発現低下とレトロトランスポゾンの活性化がどの様な関係にあるのかは明らかではないが、これらの発現変化がDmrt7欠損マウスの精子形成を停止させた原因である可能性は高いと思われる。
