Research Abstract

ウシ血清アルブミン(BSA)と2-アントラセンカルボン酸(AC)の非常に複雑な結合挙動を、UV、CLI、蛍光スペクトルそしてニトロメタン添加系における蛍光寿命測定法など、各種分光法を駆使することにより検討した。BSAには、様々な基質に対する結合サイトが多数存在し、錯形成挙動は非常に複雑であるが、ACに対する結合サイトに関して、分光学的手法を駆使することにより、明確にAC-BSA錯体組成比ならびに、結合定数を明らかにすることができた。すなわち、BSAにはACに対する独立した4つのキラルな結合サイトを有し、第1、第2、第3、第4サイトにそれぞれ1分子、3分子、2分子、3分子のACが、5.3x10^7M^<-1>、1.3x10^5M^<-1>、1.4x10^4M^<-1>、3x10^3M^<-1>の錯形成定数で結合することを明らかにした。また、蛍光スペクトルより、BSAのAC結合サイトはかなり高い疎水性を示し、特に第1サイトはペンタンと同程度の極めて疎水性の高い空間であることが明らかとなった。
さらに、ニトロメタンによるBSAに結合したACの消光実験より、各サイトの消光速度定数を求めた結果、第1サイトは約32倍、第2サイトは約8倍、水中のACよりニトロメタンによる消光を受けにくく、第3、第4サイトは水中のACと同程度の消光を受けることが明らかとなった。以上の結果より、第1、第2サイトはBSA内部に存在するのに対し、第3、第4サイトは表面近傍に存在することが示唆された。これらの情報により、ニトロメタンがサイト選択的消光剤として利用可能であることを明らかとした。
上記知見を基に、BSAを不斉反応場とする、ACの超分子不斉光環化二量化反応を詳細に検討し、最高で58%eeと不斉光化学反応としては比較的高い値が得られ、BSAが超分子不斉光化学反応の有効な不斉反応場として機能することを初めて明らかにした。
