Research Abstract

1分子蛍光イメージングに用いられる対物型エバネッセント照明光学系をもとに新しいレーザー暗視野光学系を開発し、回転分子モーターF_1-ATPaseの高時間・高空間分解能計測に適用した。
構築した計測システムの光学系は、従来の全反射型蛍光顕微鏡(エバネッセント顕微鏡)に用いられるものとほぼ同じである。新システムでは、ダイクロイックミラーの代わりに中心部が光を透過するミラーを配置しているだけである。試料を照明するレーザー光はミラーで反射され検出器(Hi-speed camera)には入らないが、試料由来の散乱光はミラーの中心部を通って検出器に投射され暗視野像が得られる。照明光を励起光としても利用すれば,暗視野像と蛍光像を同時に取得することも可能である。さらに通常の暗視野顕微鏡と異なりコンデンサーレンズを必要としないので試料上部にスペースを確保でき、マイクロマニピュレーターや細胞培養装置等との併用もできる。直径40nmの金コロイドをプローブとした際には、時間分解能9.1μs(使用した検出器の限界値)で1.5nm程度の位置決定精度が達成でき、従来の暗視野顕微鏡を用いた以前の報告例(320μsで5~6nmの精度)を大幅に改善できた。さらにこの計測システムをF_1モーターの回転計測に適用した結果、回転停止中のプローブの揺らぎからF_1の涙じり弾性(剛性)が求められた。また回転中のF_1の角速度は一定にならないことが明らかとなり、この結果からF_1のトルクには角度依存性があることが示唆された。
