無標識蛍光相関分光による生体高分子のナノ秒揺らぎの研究

Research Abstract

本年度は、以下に述べる研究を行った。
1.二光子励起蛍光を用いた紫外光吸収色素の蛍光相関分光の試み
前年度までに準備した光学系および試料交換系を用いて紫外部に吸収を持つトリプトファン等の二光子励起蛍光相関測定を検討したが、フローセルによる試料交換のみでは褪色の影響を効果的に除くことに限界があり、現時点では天然蛍光物質を利用したナノ秒に至る時間分解能での蛍光相関測定は困難であることが分かった。
2.マイクロ流路セルを用いたミリ秒-フェムト秒二次元時間分解蛍光分光装置の開発
前項の目的のために購入したマイクロ流路セルとシリンジポンプを活用し、既存のフェムト秒時間分解蛍光分光装置を拡張して生体高分子の高次構造形成中間体(ミリ秒オーダー)を追跡するための装置を製作した。これを用いて、pHジャンプに伴うヒト血清アルブミンの構造変化を蛍光プローブ分子周囲の水和ダイナミクスの変化を通して観測することができた。
3.蛍光寿命揺らぎを観測する蛍光相関分光法の生物学的問題への応用
独自に開発した蛍光寿命揺らぎを観測する蛍光相関分光法を応用し、ヘアピン構造を取る一本鎖DNAの構造揺らぎをFRET(蛍光共鳴エネルギー移動)による蛍光寿命変化を指標として調べた。蛍光寿命の揺らぎとして観測された約100マイクロ秒のダイナミクスをシミュレーションと比較した結果、3つの異なる状態を仮定することでこれを再現できることが分かった。このモデルは以前に提出された3状態モデルと定性的に一致しないものであった。
