Research Abstract

細胞内ロジスティクス機構と歯周病菌と細胞侵入の関連について検討を加えた。歯周病菌Porphyromonas gingivalis(Pg)は歯周細胞に侵入する.歯肉上皮細胞内に侵入したPgはまず初期エンドソームのマーカーであるFYVEと共局在を示し,その局在は経時的に減少した.その後,細胞内のPgの約半数がLAMP1(ライソソームマーカー)と共局在を示した.また,同じく感染1時間後,細胞内のPgの約70%はトランスフェリンレセプターと共局在を示し,その局在は経時的に減少した.このことから,歯肉上皮細胞内に侵入したPgはまず初期エンドソームに存在し,ライソソームで分解を受ける菌がいる一方,エンドサイトーシス経路へとソーティングされる菌がいる可能性が示された.また,エンドソームから細胞膜へのリサイクリングを制御するRabファミリーGTPaseであるRab11,およびアクチン細胞骨格系の再構成を制御するGTPaseであるRalAとPgとの共局在が観察された.RabllとRalAのドミナントネガティブ型(Rabll^<25N>,RalA^<27N>)とPgとの共局在を検討したところ,野生型と比較し,その共局率は減少した.さらに,RabllとRalA遺伝子のRNAiノックダウンにより,細菌の細胞培養液中への脱出の減少と細胞内の生菌数の増加がみられた.さらに,リサイクリング小胞の細胞膜への繋留因子であるエキソシスト複合体の1つであるSec5,Sec6およびExo84遺伝子のRNAiノックダウンを行ったところ,細胞培養液中の生菌数の減少,並びに細胞内の生菌数の増加がみられた.
これらの結果より,歯肉上皮細胞に侵入したPgの一部はRabllとRalAが制御するリサイクリング経路ならびにエキソシスト複合体利用して細胞外へ脱出し,さらに近接細胞へ再侵入し,歯周組織の感染拡大を果たしていることが示唆された.
