Research Abstract

本研究では,細胞内での核の3次元形態や,核のひずみ,核の力学特性を精密に計測し,核に生じる力を求める手法を確立していくとともに,力の要因となる核内外の要素を明らかにしていく.そして,細胞の分化の違いなどによって,核の形態や核内の応力場がどのように変化していくのか,明らかにしていくことを目的としている.
本年度は,特に細胞内張力を負担していると考えられるアクチン細胞骨格に注目し,これらが細胞核の力学場に与える影響を詳細に調べた.すなわち,現有する細胞骨格切断用のレーザアブレーションシステムを用いて生細胞内の個々のストレスファイバを切断する,あるいは,薬物処理によって選択的にストレスファイバを破壊する,などして,ファイバの張力を解放したときの細胞核の挙動を詳細に調べた.この過程で,細胞内の核には,アクチン細胞骨格を主体とした細胞内張力に起因する圧縮力が加わっており,細胞内張力変化に応じて核の3次元形態が可逆的に変化することなどを見いだした.さらに,わずか1本のストレスファイバの収縮力によって,ファイバ直下の核に局所的に変形が生じるという結果が得られた.これらことから,細胞骨格と核膜との間には,かなり大きな力学的インタラクションが作用していることが予想された.
