スモ修飾システムによる修復関連核内ドメイン形成制御機構の解明

Research Abstract

本研究では、タンパク質翻訳後修飾のひとつであるスモ修飾システムに注目して、DNA二本鎖切断の相同組換え修復機構にける修復関連核内ドメイン形成における役割の検討を進めた。まず、紫外線マイクロ照射法を用いてゲノム損傷部位へのスモの集積を検討したところ、損傷誘導5分後からスモタンパク質のひとつであるスモ1とスモ化酵素UBC9が集積することが確認され、スモ修飾システムがゲノム修復機構に関与が示唆された。スモタンパク質と相互作用する修復関連蛋白質としてはRAD51が知られている。そこで、RAD51の損傷部位への集積にスモ修飾システムが関与しているかどうかの検討を行った。siRNA法を用いてスモ1の発現抑制を行ったところ、紫外線マイクロ照射法によるRAD51の損傷部位への集積が抑制され、スモ修飾システムのRAD51フォーカス形成への関与が示唆された。さらに、スモ修飾システムのRAD51動態制御への関与は、SUMO化酵素UBC9やPIAS4の発現抑制細胞を用いた紫外線マイクロ照射法によるゲノム損傷部位へのRAD51集積抑制により確認された。RAD51の損傷部位への集積、すなわちゲノム修復関連核内ドメインの形成には、ゲノム損傷応答により活性化されたスモ修飾システムが関与していることが明らかになった。現在、これらのデータをまとめて、論文投稿中である。
