Research Abstract

我々は、これまでにショウジョウバエ幼虫を使用した嗅覚連合学習パラダイムを確立し、キニーネ罰学習では最長20分の短期記憶しか形成されないが、ショ糖報酬学習では2時間にわたり維持されるCREB依存性中期記憶が生成されることを明らかにし、条件刺激と非条件刺激を統合する神経回路の違いが記憶の安定度の違いを生み出す主要な基盤である事を提唱してきた。この仮説を検定するために、嗅覚受容体遺伝子プロモーター調節下でチャネルロドプシン(ChR2)を発現させ、匂い刺激を青色光刺激に置きかえるさまざまなショウジョウバエ系統(Or82a-ChR2,Or42b-ChR2,Or47a-ChR2,Or24a-ChR2,Or83b-ChR2)を作製した。さらに、ショ糖報酬刺激を温度刺激に置きかえるために、オクトパミン神経特異的なTDC2遺伝子のプロモーター調節下で温度依存性チャネルdTrpA1を発現させる系統(TDC2-dTrpA1)を作製した。また、キニーネ罰刺激を温度刺激に置きかえるために、ドーパミン神経特異的なTH遺伝子プロモーター調節下で温度依存性チャネルdTrpA1を発現させる系統(TH-dTrpA1)を作製した。少なくとも5回の戻し交配によりこれらの系統の遺伝学的背景を標準化した後に、構築した系統の掛合わせにより様々なOr-ChR2とTDC2-dTrpA1またはTH-dTrpA1の両者を持つショウジョウバエを構築した。幼虫行動解析により、構築した系統は青色光または温度刺激により誘引される事を明らかにし、これらの人為的神経刺激が外部刺激を期待どおりに置換する事を確認した。さらに、1)温度刺激と青色光、2)青色光のみ、3)温度刺激のみの条件付けをおこない、人為的刺激による連合学習の効果検討を進めている。
