Research Abstract

精子幹細胞ニッシェは精細管の基底膜と支持細胞であるセルトリ細胞により構成されている。本研究ではニッシェ制御に関わると見られる分子の解明を目的としている。本年度の研究ではG蛋白質分子のRacがニッシェへの幹細胞のホーミングに関わる役割を調べた。Quantitative RT-PCRにより精巣から抗EPCAM抗体を用いたMagnetic sortingにより濃縮した精原細胞集団および精子幹細胞の長期培養系(Germline stem : GS細胞)についてRacの発現を調べたところ、Rac1が強い発現を示すことが分かった。
Rac1の発現はGS細胞のlamininへの接着によって亢進し、一方GS細胞の増殖促進因子であるFGF2やGDNFにより低下した。そこでRacのDominant active(DA)およびDominant negative(DN)を強制発現させたGS細胞を樹立した。DA-Rac細胞とDN-Rac細胞はいずれも精原細胞マーカーの発現に野生型と差異が見られなかったが、DN-Rac細胞にて増殖やラミニンへの接着の亢進が認められた。さらにDN-Rac細胞は精巣内移植によりコロニー形成率が亢進し、またserial transplantationにより自己複製活性も亢進していることが分かった。またホーミング現象を試験管内で再現するため、セルトリ細胞をフィーダーとした培養系の開発を行った。
