Research Abstract |
1.植物ROS生成酵素rbohの網羅的解析 シロイヌナズナrboh遺伝子の発現部位の解析と、異種発現系を用いたROS生成活性の解析を並行して進め、両者を総合して、植物体での各遺伝子の機能分担について考察した。 AtrbohH,Jは、花粉に特異的に局在することが明らかになったので、二重突然変異体等を用いて、rbohを介したROSの局所的生成が花粉管の発芽、先端成長の制御に関与している可能性を検証した。 2.rbohの活性制御因子の単離と機能解析 2-1:異種共発現系を用いた活性制御候補因子Racによるrbohの制御機構の解明 シロイヌナズナの11種全てのRac遺伝子をクローニングし、Atrboh遺伝子との異種発現系における共発現解析により、ROS生成量を調べた。 2-2:酵母two-hybrid法を用いた植物rboh新奇活性制御候補因子のスクリーニング rbohの活性制御にはCa^<2+>の結合とリン酸化以外にも、特にN末端細胞質領域において複数の制御因子との相互作用が関与する可能性がある。そこで、感染防御応答や気孔閉鎖等に関与するAtrbohD,AtrbohFのN末端領域と相互作用する因子を酵母two-hybrid法により単離を試みた.これまでに,相互作用候補因子としてプロテインキナーゼ,防御応答関連因子を含む19種を単離した異種共発現解析系を用いて、rbohのROS生成活性に対する候補因子の共発現の影響について検証した。
