Outline of Research at the Start

幹細胞は、特定のヒストンバリアントを取り込むことで特異的なクロマチン構造を形成し、通常の体細胞より全遺伝子の転写活性状態が極めて高い高転写状態にある。中でも成熟卵母細胞、精原細胞の胚性前駆細胞である始原生殖細胞の解析は最も先駆的に行われている系の一つである。高転写状態は、初期発生時の爆発的な細胞増殖や多系統への分化に不可欠と考えられているが、その分子機序はいまだ不明である。高転写状態がもたらす遺伝子発現の“量的”制御の解明にはクロマチン構造の形成による転写量制御から、タンパク質の発現まで包括的に理解する必要がある。そこで、本研究では、新規技術開発を行う。

Outline of Annual Research Achievements

私たちはこれまで免疫染色をベースに、細胞・組織を非破壊的に標的タンパク質を可視化し、同時にゲノム上の位置を決定するChILSeqを開発した。ChILSeq では抗体反応領域周辺のゲノムDNAにTn5トランスポザーゼを用いて選択的にT7RNA ポリメラーゼプロモータ配列を挿入し、T7RNAポリメラーゼの添加により強制的に転写させることでゲノム情報をRNAとして獲得する。ChIL反応により転写されたRNA はゲノムから遊離しないため、細胞の形態や組織内の位置情報を保持した状態でsmFISH により検出可能である。そこで、本研究ではChILsmFISH により、NGSなしでのイメージングのみでハイスループット且つ1細胞レベルの高解像度エピゲノム解析技術が樹立し、エピゲノムとトランスクリプトームの同時測定技術の樹立を試みた。加えて、高転写状態解析に必要となるタンパク質の発現情報の取得を進めた。本研究では、プロテオミクスまで含めたマルチオミクス技術を開発にとりくんだ。プロテオミクスの適用の為新たに開発した連続蛍光免疫染色法(seqImmuno-Fluo:以下seqIF)の実装を行い、200種を超えるプローブの作成と検証を行い、ヒト検体を用いた実データ取得を進めた。更に、これら解析を行うための自動化デバイスの開発に取り組み、共焦点レーザー顕微鏡下でデータを自動的に取得し、反応を行う新たなデバイス開発に成功した。現在論文投稿準備中である。また、トランスクリプトーム技術seqFISH+との統合を進めた。
