Outline of Research at the Start

現代の時間生物学の基盤を創ったユルゲン・アショフは、ヒトの体内には自律的に約24時間周期で振動する時計、「概日時計」の存在を発見し、体温と睡眠・覚醒の概日リズムが乖離する事を報告した。彼は、ヒトの時間感覚は体温リズムではなく、睡眠・覚醒のリズムに影響を受ける事を提示した。これらの事は、ヒトの“時間”は体温調節系と睡眠・覚醒調節系の二種類存在する事を示す。しかし、これら二つの時間調節に関わる神経経路はほとんど明らかにされていない。本研究では中枢概日時計を起点とする、睡眠・覚醒リズムと体温リズム調節に関わる神経経路を明らかにし、ヒトが持つ異なる二つの時間の存在意義に迫る。

Outline of Annual Research Achievements

現代の時間生物学の基盤を創ったユルゲン・アショフ(故)は、ヒトの体内には自律的に24時間周期で振動する時計、「概日時計」の存在を発見し、体温と睡眠・覚醒の概日リズムが乖離する(異なる周期を示す)事を報告した。これを内的脱同調と呼ぶ。その中で、ヒトの時間感覚は睡眠・覚醒のリズムに影響を受け、体温リズムには影響を受けないことを提示した。つまり、ヒトの“時間”は体温調節系と睡眠・覚醒調節系の二種類存在し、それぞれ異なるメカニズムで生体の時間が調節されていると言える。哺乳類の24時間周期の概日リズムは、視床下部の視交叉上核により調節されていることが知られている。しかし、さまざまな生理機能の時間的調節が、どの経路を介して行われているかは、今日に至るまでほとんど理解が進んでいなかった。これらの背景から、申請者は視交叉上核からの出力経路の探索を進め、最近視交叉上核と睡眠・覚醒を繋ぐ、新しい神経回路を同定し報告した (Ono et al., 2020 Science Advances)。本研究では視交叉上核を起点とする、睡眠・覚醒リズムと体温リズム調節に関わる神経経路を明らかにし、ヒトが持つ異なる二つの時間の存在意義に迫る。
神経回路の機能を明らかにするためには、どの伝達物質がどのタイミングで放出され、その結果神経活動が変化しているかを理解する事が重要な点となる。そこで、ウイルスベクターを用いたin vivoゲノム編集を用いて、特定の細胞の特定遺伝子を欠損させ、神経回路に決定的な分子を同定する。また、実際の伝達物質の分泌が、どのように下流細胞の活動に影響を与えているかを理解する為、Genetically encoded GPCR Activation-Based (GRAB) センサーを導入する(Dr. Yulongとの共同研究)。視交叉上核の主な神経ペプチドであるVIP, AVPのGRABセンサーを用い、光を用いてペプチド分泌計測が可能である事を見出した。
