Outline of Research at the Start

本研究では、以前我々が見出した「水素結合型塩基対⇔金属錯体型非天然塩基対」の配位駆動型塩基対スイッチングに基づき、DNA二重鎖の組み換え(DNA鎖交換反応)によって機械的な動きが誘起されるDNAナノ構造体(DNAロタキサン・分子ピンセット等)を構築する。DNA構造体に各種機能分子を修飾し、金属錯体形成や酸化還元を駆動力としてダイナミックな構造・エネルギー変換を引き起こす「金属配位駆動型DNA発動分子」を創製する。

Outline of Annual Research Achievements

本研究は、金属錯体形成を駆動力として構造変換を引き起こす「金属配位駆動型DNA発動分子」の創製を目的とした。本年度は主に、金属イオンの有無により「水素結合型塩基対⇔金属錯体型非天然塩基対」の塩基対スイッチングが可能な新規修飾ウラシル塩基を開発し、DNA二重鎖の組み換え反応への応用を検討した。具体的には、5位にイミノ二酢酸配位子を導入したN,N-ジカルボキシメチル-5-アミノウラシル(dcaU)塩基を設計・合成した。合成したdcaUヌクレオシドをDNA鎖中に導入し、DNA中での金属錯体形成を検討した。種々の金属イオンを検討した結果、一対のdcaU-dcaU対を含むDNA二重鎖は金属錯体型dcaU-Gd(III)-dcaU塩基対の形成により大きく安定化され、水素結合型dcaU-A塩基対を含む二重鎖はGd(III)イオンの添加により不安定化されることがわかった。これは、dcaU-Gd(III)-dcaU塩基対とdcaU-A塩基対の相互変換が可能であることを示唆する結果である。さらに、dcaU塩基を含むDNA鎖を用いて、Gd(III)イオンの添加・除去による二重鎖形成の制御を試みた。Gd(III)非存在下ではdcaU-A塩基対を含む二重鎖が形成し、Gd(III)イオンを添加するとdcaU-Gd(III)-dcaU塩基対を含む二重鎖が優先的に形成した。すなわち、Gd(III)イオンを外部刺激として、DNA鎖の会合挙動を等温下で可逆的に変換することができた。昨年度までに検討した5-ヒドロキシウラシル(UOH)や5-カルボキシウラシル(caU)では、金属イオンによる二重鎖形成制御にはDNA配列中に3~4個連続して導入する必要があった。一方、dcaU塩基は一対のみでも良好な二重鎖会合制御ができたため、DNA発動分子の有用なビルディングブロックとなると考えられる。
