Outline of Research at the Start

体内“細胞外液”とその調節能を獲得したパイオニア、扁形動物の原始“抗利尿ホルモン系—腎臓”を橋頭保に、海から新しい水・塩分環境への動物の不連続な進出をもたらしたシンギュラリティを検証する。
①プラチトシン系の発現様式の解明とシンギュラリティ細胞である可能性の究明
②シンギュラリティとしてのプラチトシン系-原腎管の細胞・分子基盤の解明
③プラチトシン系-原腎管の環境進出におけるシンギュラリティ機能の個体レベルの解明

Outline of Annual Research Achievements

“プラナリア”など扁形動物は、基盤的な左右相称動物で、初めて本格的に新しい水・塩分環境に進出した。我々は、広塩性のヒラムシにおいて、抗利尿ホルモン系の祖先型を同定し、プラチトシン系と命名した。そして、抗脱水に必須であることも見出した。『抗利尿ホルモン系による“腎臓” i.e., 体液の調節』が扁形動物まで遡れ、新環境進出のシンギュラリティの可能性がある。また、プラチトシンは脳神経節の2対のニューロンのみで産生されることも発見した(シンギュラリティ細胞?)。この2対の細胞は、脊椎動物の2群に分化している数千の抗利尿ホルモン産生細胞(脳内で作用するもの・ホルモンを血中に放出し全身に作用するもの)に対応する原型の可能性がある。これらは前左右相称動物のゲノム等には見出せない。神経中枢化がはじまり“細胞外液”を獲得した扁形動物の神経ペプチド群は、神経シナプス系からの内分泌系/液性調節の誕生:神経内分泌~脳による生体制御系のシンギュラリティとも考えられる。
そこで、SPring-8での3Dマイクロ-CT法により、脳神経節を3D再構築した。前口動物(キノコ体)・後口動物(視交叉)双方の特徴をもち“原始脳”である可能性が示された。さらに、この脳神経節から、進化的に普遍的な神経ペプチドの祖先群として、生殖腺刺激ホルモン放出ホルモン(GnRH)等も発見した。現在、それらのはたらきと、1細胞/空間トランスクリプトームにより、そのニューロン回路/分子基盤を検討し、データ駆動型の発展も目論んでいる。
