Outline of Research at the Start

本研究では、生体内で「弱い相互作用」によってタンパク質とナノマテリアルなどが形成する複合体立体構造や結合親和性を予測し、結合がもたらす効果を予測する先端的な分子シミュレーションを開発する。これをリガンドやナノマテリアルと細胞表面受容体などとの「弱い相互作用」の研究に応用することで、先端的な分子シミュレーション法をマテリアル・シンバイオシスに活用可能にし、相互作用の基礎的な知見を得ることを目指す。

Outline of Annual Research Achievements

前年度に引き続き、複合体立体構造から解離を行うdPaCS-MD/MSM法、解離状態から結合・解離サイクルを繰り返すa/dPaCS-MD/MSM法やColDock法、evERdock法をタンパク質とナノマテリアルの複合体などの立体構造予測にも利用できるようにシミュレーション法やモデルの開発・改良を行った。領域内の共同研究を中心として様々な分子複合体を対象に研究を展開した。具体的には、ヘルペスウイルスのglycoprotein BとPILRαの相互作用、物質共生に大きくかかわるPEG-抗PEG抗体の弱い相互作用、味覚受容体のアミノ酸認識、SGLT1による糖の選択的認識、アデノシン A2A 受容体と低分子化合物の相互作用などの研究を領域内の共同研究者の実験データと比較しながら研究を展開した。glycoprotein BとPILRαの複合体やSGLTと糖の相互作用に関しては、先行研究で指摘されていた分子力場依存性が我々の研究でも確認でき、これを踏まえた研究を行った。SGLT1に関しては、当初実験で適当な立体構造が見られなかったため、SGLT2の実験構造からホモロジーモデリングを行ったが、年度中に実験構造が得られたのでこれに基づく再計算を行った。PEG-抗PEG抗体の相互作用に関しては、九大森研究室との共同研究でTI抗原とTD抗原における違いを明らかにし、この結果を含む論文の執筆を進めている。味覚受容体のアミノ酸認識においては、2種類の類似したアミノ酸の結合の強さの違いをもたらす原因を明らかにすべく計算を行った。これらの場合はPaCS-MDを使わなくてもシミュレーション時間内でしばしば解離や再結合が見られたので、多数のMD計算を実行し更にMSM解析を行うことで、結合と解離の過程の詳しい解析を行うことができた。
