Outline of Research at the Start

高時間分解能・単一分子力学測定技術を用いて、分子の会合~結合形成~解離の一連のプロセスをナノ秒スケールで1分子レベルで解析する。これにより、微視的に観察される分子会合・結合形成ダイナミクス(ピコ~ナノ秒)と巨視的に観察される結合キネティクス(秒~時間)を双方向に議論可能な学理を構築する。さらに、生体環境に代表される分子夾雑状態の結合キネティクスへの影響を、微視的分子シミュレーション、巨視的反応速度論で定量的に取り扱うための方法論を確立する。

Outline of Annual Research Achievements

本研究では、以下の3つを達成した。1.制御された分子夾雑状態での生体分子間、生体分子-リガンド間の接近-結合形成-解離のプロセスにおける相互作用力の、ナノ秒スケールの時間分解能でのリアルタイム計測。2.ナノ秒スケールの微視的分子ダイナミクスと秒~時間で観測される巨視的な反応次数・速度定数の相関解析。3.微視的な分子ダイナミクス巨視的キネティクスにおける分子夾雑環境の影響の定量的表現技術の確立。
1では、原子間力顕微鏡と高速デジタイザを組み合わせる手法を開発し、1 nm/秒という極端に遅い速度で分子間の位置を変化させつつ、分子間相互作用を高速サンプリングした。その結果、ストレプトアビジン-ビオチン分子間の相互作用を、10 pN以下の応力分解能、500ナノ秒の時間分解能で解析することに成功した。これにより、過渡状態や非特異的相互作用などによる複数の準安定状態の存在が明らかになり、カンチレバーの位置、変位から結合ポテンシャル地形の全体像を得ることが可能となった。
さらに時間分解能を向上させ、100ナノ秒以下の時間分解能で結合力のダイナミクスを精密計測した。これにより、1分子ダイナミクスの分子シミュレーションと実験の結果の直接比較が可能となり、分子結合形成ダイナミクス(分子の試行錯誤)の詳細に迫ることができた。これらの研究成果は、微視的な分子ダイナミクスと巨視的キネティクスとの間の相関解析、分子夾雑環境の影響の定量的表現技術の確立といったにも貢献する可能性があると考えられる。
