Outline of Research at the Start

本研究の目的は、凍結保護活性やタンパク質の凝集抑制活性を持つ保護高分子化合物と、細胞膜やタンパク質との弱い相互作用による保護活性のメカニズムの解明とそれを応用した分子設計である。細胞膜やタンパク質に覆い被さるような形で保護するというMolecular Shielding効果を、分光学的手法や熱測定により定量的に解析し構造活性相関を明確にする。生物が持つ特殊な耐乾燥性保護ペプチドなどとの活性と構造を比較し、生物及び化学の両面からアプローチすることで生体保護材料という新たな物質共生の新展開を目指す。

Outline of Annual Research Achievements

凍結保護高分子およびタンパク質保護高分子のメカニズム解明に向け、細胞膜およびタンパク質との弱い相互作用を測定し、Molecular Shielding仮説の検証を行った。
双性イオンポリマーに対して疎水性部位を導入することにより、高分子とタンパク質および高分子と細胞膜リン脂質との相互作用が強まることを等温滴定型カロリメトリーにより、定性的に調べた。定量的な議論のため、Pulsed Field Gradient-NMR法により、タンパク質およびリン脂質に対して双性イオンポリマーを添加することで、それらの拡散係数の低下から相互作用を見積もった。その結果、疎水性相互作用による弱い相互作用によるものであることが確認された。次に、分子動力学シミュレーションにてインスリンと双性イオンポリマーの相互作用を評価したところ、疎水性部位を導入することでインスリン分子間に可逆的に挿入されることで、インスリン同士の凝集を抑制していることが示される結果を得た。これらと、実際のタンパク質凝集抑制試験の結果を総合的に判断し、Molecular Shielding仮説の有効性を議論することに成功した。これらの成果は計7本の論文にて発表した。
