Outline of Research at the Start

統合失調症や自閉症のリスクとして、母胎の精神的ストレスと子のGABA関連遺伝子の異常には証拠がある。そこで遺伝-環境交互作用に着目して、GABA合成酵素 GAD67遺伝子Gad1のヘテロ欠損胎仔の母体に拘束ストレスを加えるTwo-hit モデルを作製した。このモデルはparvalbumin (PV)陽性GABA細胞の減少と行動・脳波の異常を表出し、DNAメチル化変化によりGABAから acetylcholine (ACh)へと細胞発生・分化が運命転化した可能性がある。そこで、GABA/ACh細胞発生転写因子発現の臨界期にエピジェネティクに誘導されたACh細胞によって規定のGABA 神経回路が新奇に更新されるメカニズムを解明する。

Outline of Annual Research Achievements

1.臨界期ストレス暴露大脳皮質錐体細胞の電気生理学的解析:大脳皮質内側前頭前野スライス標本を作製し錐体細胞からパッチクランプ記録を行い、抑制性電流の変化を対照群と比較した。両群ともにドネペジル投与後にsIPSCの振幅が増加し、アトロピンで対照群は元に戻ったがストレス群ではsIPSCの振幅がさらに減少した。sIPSCの頻度はドネペジル投与後は対照群で頻度が増し、アトロピン投与後対照群は元に戻ったが、ストレス群ではさらに頻度が減少した。強化されたGABA作動性抑制性神経伝達がAChの放出増加とムスカリン受容体によることを同定した。
2.臨界期ストレス暴露成体大脳皮質GABA 細胞の電気生理学的機能解析:同様にスライス標本を作製してGABA 細胞からパッチクランプ記録を行った。ストレス群では基電流減少、後過分極電位減少、入力応答発火頻度の増加がみられ、これらの変化はアトロピンで消失した。GABA細胞において増加したACh神経終末から放出されたAChにより、GABA細胞のACh感受性K電流(M電流)が抑制を受けと考えられた。ニコチン性受容体阻害薬のMLAでは若干の、DhβEでは同様の傾向があったが、実験手技上アトロピン作用が残存していた可能性を否定できなかった。
3.臨界期ストレス暴露成体大脳皮質の神経回路機能解析:臨界期ストレス暴露によるACh入力の増加とその影響を明らかにするために、対照群とストレス群にドネペジルあるいはアトロピンを腹腔内投与して、ストレス群における脳波のγ帯域の減少や行動テストの社会的新規性認識の障害が改善するかどうか検討した。
