Research Abstract

胚発生において、細胞の発生運命と細胞分裂回数は深く関わっていることが知られているが、本研究では、細胞の発生運命の決定・細胞分化と、細胞分裂停止までの細胞分裂回数との関係を、ホヤ胚を用いて解析している。
胚発生において、細胞の運命決定がなされた直後から組織特異的なキー転写因子群が発現するようになる。脊索で発現が開始するBrachyury遺伝子は、脊索分化を司るキー転写因子であることがわかっていた。昨年までの本研究によって、Brachyuryは脊索細胞が64細胞期以降3回で細胞分裂を停止するために必要かつ十分であることを示してきた。そして、本年これらをまとめた論文がDevelopmental Biologyに出版された。
そこで、64細胞期以降3回で細胞分裂を停止する筋肉細胞にも同様のことが起こるのかどうかを調べた。筋肉における組織特異的なキー転写因子はTbx6であり、これは筋肉分化に必要かつ充分である。よって、Tbx6 mRNAを合成し、受精卵に顕微注入をおこなった。その結果、64細胞期以降7回の細胞分裂を行うはずの間充織割球が、筋肉同様に3回の分裂後に停止することがわかった。このように、脊索と筋肉の両方に共通して、組織特異的なキー転写因子は分化と分裂回数制御の両方に関わっていることが示された。しかし、Tbx6 mRNAの顕微注入によりTbx6タンパク質を正常より早い時期に発現させているにもかかわらず分裂回数が3回と変わらなしでことから、Tbx6遺伝子の発現開始時期からの時間で分裂回数が決まっているわけではないことがわかっていた。
