Research Abstract |
細胞分裂において中心体は微小管ネットワークの再編のかなめとして機能し、紡錘体形成に重要な役割を果たしている。なかでもAurora-A、Plk1などのmitotickinaseは微小管ネットワークの再編の制御因子として機能している。一方で、分化終末細胞である神経細胞は細胞分裂に向かうことはないがmitotickinaseは多く発現しておりその機能は不明であった。我々は後根神経節細胞を用いた実験からAurora-Aが神経細胞の神経突起伸展の際の微小管ネットワーク再編に必須であることを証明した。さらに、Aurora-Aの標的タンパク質であるNDEL1が協調して機能することを明らかにした。また、我々はAurora-Aが神経突起の進展だけでなく、発生期における神経細胞の遊走にも関与することを明らかにした。小脳顆粒神経細胞を用いた遊走実験から、遊走開始と同時にAurora-Aのリン酸化が亢進し、微小管の中心体からの進展が促進されることが分かった。逆に、Aurora-Aのノックダウン、阻害剤で抑制した時には有意に神経細胞の遊走が阻害されることが分かった。これらのことはmitotickinaseは細胞分裂だけでなく分化終末細胞においても積極的な機能があることが分かった。さらに、CDK5はAurora-Aの上位のリン酸化酵素として機能し、CDK5rap2を介してAurora-Aの中心体への集積を制御し、両者はNdel1を介して微小管のダイナミックスを制御し、神経細胞の遊走をコントロールしていることを証明した。
