Research Abstract

現在までに、RNA結合タンパク質Khd1によるmRNAの安定性と翻訳制御の分子機構とその生理的意義について研究をすすめている。出芽酵母のRNA結合タンパク質であるKhd1は、ヒトのRNA結合タンパク質であるhnRNP Kの酵母オルソログである。Khd1はHO遺伝子のリプレッサータンパク質をコードするASH1 mRNAの翻訳抑制や、膜タンパク質をコードするMTL1 mRNAの安定性制御に関与する。Khd1は.ASH1 mRNAやMTL1 mRNAを含む数百種以上のmRNA群と相互作用することが報告されているが、ASH1 mRNAとMTL1 mRNA以外の標的mRNAに対するKhd1pの作用は不明であった。また、Khd1Δ単独変異株の増殖速度は野生型株と同程度であり、Khd1pが多種類の標的mRNAと相互作用する生理的意義も不明であった。今年度、Khd1がポリA分解酵素Ccr4,Caf1とオーバーラップした機能をもち、出芽酵母の芽の局所的な細胞壁合成と維持(Cell wall integrity)の制御に関与することを明らかにした。また、Khd1の新たなターゲットとして、ROM2遺伝子を見出した。さらに、Khd1とCcr4による細胞壁合成の維持の経路に、RNAヘリカーゼDhh1が関与することも明らかにした。これらの成果の一部は論文発表しました。さらに、Khd1がシグナル認識粒子Signal recognition particle(SRP)と結合すること、SRPとポリA鎖分解酵素Caf1が結合すること、SRPとRNAヘリカーゼDhh1と遺伝学的相互作用があることなどを見出しており、Khd1によるmRNAの安定性と翻訳制御の分子機構について、さらなる研究をすすめている。
