Research Abstract

【平成23年度】Atg32-Atg8および-Atg11間相互作用とAtg32機能ドメインの機能解析
Atg32は、ミトコンドリア・オートファジーの選択性を規定する鍵タンパク質であり、オートファジー関連因子Atg8およびAtg11と直接相互作用する。この相互作用がミトコンドリア分解に重要であることを、それぞれの相互作用に特異的な異常が見られる変異タンパク質を解析することにより、平成22年度までに明らかにした。引き続き今年度において、Atg32-Atg8およびAtg32-Atg11の相互作用が、オートファジー欠損によって影響を受けないことを見出した。このことは、ミトコンドリアを隔離するオートファゴソーム膜の形成に依存せずに、Atg32はAtg8やAtg11と複合体を形成すること、この複合体形成がマイトファジーの初期の段階で起こっていることを示唆している。
また、Atg32がマイトファジーを誘導する培養条件でリン酸化されることを明らかにした。Atg11との相互作用に異常を示すAtg32変異タンパク質はリン酸化を受けないことから、リン酸化がAtg32-Atg11相互作用を制御している可能性が考えられる。加えて、オートファジーに必須なタンパク質キナーゼAtg1がAtg32のリン酸化に関与していること、Atg32とAtg1が免疫共沈降することを見出した。Atg1はAtg11と相互作用することが知られており、Atg1はAtg11を介して、Atg32と相互作用していると考えられる。
Atg32の機能ドメインについては、昨年度までの研究により、ミトコンドリア外膜に挿入される膜貫通ドメインのN末側の細胞質ドメインにマイトファジーを起こす活性があることを示したが、この細胞質ドメインをペルオキシソームにアンカーすることで、ペルオキシソームの分解を誘導されることを見出した。このことは、Atg32がマイトファジーに必要かつ十分なミトコンドリア因子であり、その活性は他のオルガネラの分解をも誘導しうることを意味しており、Atg32は選択的ミトコンドリア分解を直接媒介するタンパク質であること示唆している。
