Research Abstract

骨髄系樹状細胞の初期エンドソームに局在するTLR3は、ウイルス由来のdsRNAを認識し、炎症性サイトカインやIFN-α/β産生などの抗ウイルス応答を惹起する。しかし感染細胞内でdsRNAが検知されないnegative-strand virusの感染において、TLR3依存的な炎症性サイトカイン産生がウイルス感染を増悪させることが報告され、TLR3が認識するRNA構造は多様である可能性が示唆されている。ポリオウイルス感染では細胞内に(+)鎖ssRNAとdsRNAが検出され、骨髄系樹状細胞からTLR3依存的にIFN-α/βが産生されることから、本研究では、ポリオウイルス由来のRNAを用いてTLR3を活性化するRNA構造の同定を行った。ポリオウイルスのcDNAをテンプレートに、500~1800bpの断片化された(+)鎖、(-)鎖のvirus ssRNA、それらのdsRNAをin vitro転写で作製し、TLR3活性化能をIFN-βpromoterの活性化で調べた結果、dsRNA以外にある種のssRNA断片がTLR3を活性化することが明らかとなった。この機能性ssRNAは分解に抵抗性であり、ヒト繊維芽細胞や上皮系細胞、マウス骨髄系樹状細胞からIFN-α/β,炎症性サイトカイン産生をTLR3-TICAM-1依存的に誘導した。更に、ウイルスdsRNAアナログのpoly(I:C)同様、ラフトリン依存的に細胞内に取り込まれ初期エンドソームのTLR3と共局在する事、TLR3エクトドメインのN末、C末に存在するdsRNA結合領域を介してTLR3を多量体化させシグナルを伝達することが判明した。二次構造予測と生化学的解析から、機能性ssRNAは比較的長いタンデムなステム構造をもつことが明らかとなった。TLR3は、dsRNAのみでなく、ウイルス感染細胞から漏出した分解抵抗性のステム構造をもつRNAも感知し、シグナルを伝達すると考えられる。
