Research Abstract

【研究内容】自己の分子認識機構を解明するため,本年度はB1細胞,B2細胞におけるPIR-Bとそのリガンドによる抗体産生制御の機構の解析に注力した。
【研究結果】新たなPIR-Bのリガンドであることが示唆されたNogoについて,Nogo欠損マウスの表現型の解析を行った。またヒトB細胞集団の中で,マウスB1細胞に相当すると思われるサブセットが同定されたため(Griffin DO et al., J. Exp. Med. 2011)これに倣い末梢血B細胞集団中のB1細胞の分画,表面LILRBアイソフォーム発現の解析を行った。結果として,Nogo欠損はB細胞分化の過程と末梢での維持に影響を与えることが分かった。また「ヒト末梢血からCD20,CD27,CD43発現を指標としてごくマイナーな分画であるB1細胞の集団を分取できることが分かった。興味深いことにこの集団は前記マーカーの発現のみならずB2細胞と異なる抑制系レセプターLILRB発現プロファイルを示すことが示唆され,さらにこれが末梢血単球のLILRB発現プロファイルとも異なる傾向にあった。
【成果の意義】B細胞の制御は,非自己に効果的に反応する一方で自己への有害な反応を回避するために極めて重要である。このシステムが崩壊すると自己抗体が過剰に生産されて自己免疫疾患を惹起する。そのためB細胞は多様な制御機能を持った分子群により調節されている。自己反応性を有する血中抗体のソースであるB細胞のうち,マイナーなB1細胞集団の分画ができるようになり,B1細胞とB2細胞の抗体産生に関する抑制機構の相違を示唆する知見が得られた。PIR-B,ヒトLILRB,そのリガンドNogo等の抑制性受容体システムの調節により免疫学的な自己の確立が維持される可能性が示唆された。本研究により免疫疾患を予防,治療することも可能になる将来展開の基盤が構築された。
