Research Abstract

平成23年度は前年度までに得られた知見をもとに集積化反応をβ,γ-アルキニル-α-イミノエステルおよびグリオキシル酸由来のα-イミノエステルを基質として用い、さらに高度に集積化された反応への拡張を行った。具体的には以下の成果を得た。
(1)β,γ-アルキニル-α-イミノエステルに対して有機金属試薬を作用させることにより、N-アルキル化された中間のジエノラートを効率良く発生させ、それに対し求電子剤を作用させα-およびγ-アルキル化の位置選択性を検討し、ルイス酸の有無、およびアシル化剤の種類により位置選択性の制御が可能であることを見出した。
(2)β,γ-アルケニル-α-イミノエステルを用い、有機金属試薬によるN-アルキル化反応の検討を行ったところ、30%程度でN-アルキル化生成物は得られなかったが、基質をβ,γ-アルケニル-α-イミノチオエステルに変えたところ、N-アルキル化の収率を大幅に増加することができた。
(3)グリオキシル酸エチル由来のα-イミノエステルを用いN-アルキル化反応を行い、生じるアルミニウムエノラートに対し求電子付加反応が良好に進行することを見出した。特に添加剤として2-メトキシ-N、N-ジメチルエチルアミンを加え、アセトニトリル溶媒中-45から-30℃で6時間反応させることで目的の付加体が良好な収率で得られることがわかった。またマクロミキサーを用いたフロー合成により室温でも反応が収率よく進行することも見出した。
(4)ルイス酸を用いたアルキニルケチミンとケテンシリルアセタールとの共役付加によりイミノシクロブテノンを高収率で得られることを既に報告しているが、マイクロリアクターを用いたフロー合成により温和な条件下イミノシクロブテノンを効率よく得ることができた。
