Research Abstract

計画1)ATL31を中心とした細胞内C/Nバランサーのシグナルネットワークの解明:
C/N応答ユビキチンリガーゼATL31/6の標的として14-3-3タンパク質について検証を進め,14-3-3の安定性制御がC/N応答に関係することを証明した[Plant Journal 2011 ; Plant Signal. Behav. 2011]。ALT31C末領域のリン酸化に着目した研究を実施し,少なくとも4アミノ酸残基置換ATL31では正しいC/N応答が起こらないことを明らかにした。この結果,C/N応答にATL31のリン酸化が関与するとの示唆を得た。現在このリン酸化に関与するキナーゼの同定を進めている。
計画2)細胞内C/Nバランサーと器官間の代謝産物分配(ソース・シンク)制御に関する検討:
C/N応答の総合的理解のため,各種オーム解析を実施している。ATL31過剰発現体(ATL31ox)と欠損変異体(atl31)を用いて,C/N変動条件下におけるリン酸化プロテオーム,メタボローム解析,14-3-3タンパク質のインタラクトーム解析,網羅的酵素解析等を実施した。
計画3)高CO2条件下における植物のC/Nバランス応答・成長制御の解析:
CO2濃度調節機能を付加した人工気象器を用いて,様々な形質転換植物の高CO2応答について解析している。現在までに,通常のシロイヌナズナ野生型が高CO2生育条件下において老化の促進がみられるのに対し,ATL31過剰発現体では,それが抑制されることを明らかにした。今までの研究では,培地中の糖濃度を変化させることでC/N応答をモニターしてきたが,今後は大気中のCO2濃度を変化させることで,CO2/N応答についての解析を進める。
