Research Abstract

気孔開閉調節に関わるタンパク質PCaP1およびCO_2透過性アクアポリンPIP1に焦点を当て、生理機能の作動機構を明らかにし、高濃度CO_2下での両分子のCO_2供給システムにおける役割とそれに関わる量的・機能的調節を解明する。新規のCa結合タンパク質であるPCaP1は細胞膜に局在し、情報伝達にかかわるカルモジュリンとホスファチジルイノシトールリン酸(PtdInsPs)と結合する。PCaP1は孔辺細胞にも発現し、その遺伝子欠失株の葉では気孔の閉口ができないという表現型を観察してきた。
本年度、PCaP1が孔辺細胞に発現していることを確認し、PCaP1のタンパク質化学的な特性として、CDスペクトル解析により、Ca結合による構造変化、ITC法によるCa結合のキネティクスを明らかにすることができた。なお、気孔以外での役割の一つとして、PCaP1は病理応答にも関わっていることを明らかにすることができた。さらに、PCaP1のT-DNA挿入遺伝子破壊株では、暗所での気孔閉口が不完全となること、野生株と比べて、暗所下および高濃度CO_2条件での生育が良いことを見出した。これらの結果は、生育環境が悪いときPCaP1は生育を抑制するブレーキ役を果たしている可能性を示唆している。
多様なアクアポリン分子種の中でも細胞膜局在性のPIP1は複数の植物でCO_2透過性が認められており、申請者らはPIP1の量の減少がCO_2固定能の低下をもたらすことを見出した。高濃度CO_2に対するPIP1の応答を解析し、組織内CO_2供給システムの解明とモデルの提案を目指し、CO_2濃度の影響が高温環境で変化するか否かも解析の準備を進めた
